Spawn Cryopreservation of Agaricus Bisporus and A. Subrufescens Strains

چکیده
One of the main problems for conservation of mushroom germplasm is that the traditional method of subculturing facilitates aging and contamination of cultures. The conservation of strains at ultra-low temperature in liquid nitrogen (-196 °C) is a widely used method, however, this method become expensive in collections with large number of strains and requires highly specialized personnel. The aim of this study was to evaluate the viability of spawn of A. bisporus and A. subrufescens strains frozen at different temperatures (-20 and -80 oC). Two strains of A. subrufescens and one strain of A. bisporus were studied using spawn prepared in sorghum seeds. Fully-incubated sorghum seeds were placed in polycarbonate vials with a sterile glycerol solution (10% v/v) (G) or without glycerol (WG) and frozen at -20 or -80 oC. After 3, 6 and 12 months the samples were thawed (30 °C, 10 min) and the percentage of sample recovery was evaluated. Mycelial growth was also evaluated in potato dextrose agar (PDA) or a medium with glucose and compost extract (C) recording mycelial diameter after 7 days of incubation at 25 °C. The results showed that in the samples frozen at -20 °C, the recovery was minimal (2 to 4%). However, the samples frozen at -80 °C showed a recovery of 100% from 1 to 6 days after thawing. The recovered samples had a significantly higher growth in the medium C. In all cases the recovered samples produced normal basidiomata when cultivated on compost substrate.

Preservation of Agaricus subrufescens strains at low temperature by using cultures on sorghum grains.
BACKGROUND One of the main problems for the preservation of genetics resources of Agaricus subrufescens is to maintain the viability of the strains because the mycelium is very sensitive to cooling and therefore it ages rapidly. AIMS Evaluate the viability of A. subrufescens strains stored as cultures on sorghum grain (spawn) at different temperatures. METHODS Eighteen strains of A. subrufe...
متن کاملThe Genetic Linkage Map of the Medicinal Mushroom Agaricus subrufescens Reveals Highly Conserved Macrosynteny with the Congeneric Species Agaricus bisporus
Comparative linkage mapping can rapidly facilitate the transfer of genetic information from model species to orphan species. This macrosynteny analysis approach has been extensively used in plant species, but few example are available in fungi, and even fewer in mushroom crop species. Among the latter, the Agaricus genus comprises the most cultivable or potentially cultivable species. Agaricus ...
متن کاملجداسازی و تعیین برخی خصوصیات جهشیافتههای حاصل از پرتوتابی با اشعهی ماوراء بنفش در قارچ تکمهای سفید (Agaricus bisporus)
Abstract: The white button mushroom (Agaricus bisporus) is one of the most widely cultivated edible mushroom species in the world. Despite the long term cultivation and its economic importance, the breeding efforts in the this species have been limited. The main reason is typical life cycle that restricte cross ability between single spores of different strains. So variability in Agaricus...
متن کاملCryopreservation at -75°C of Agaricus subrufescens on wheat grains with sucrose.
Agaricus subrufescens is a basidiomycete which is studied because of its medicinal and gastronomic importance; however, less attention has been paid to its preservation. This study aimed to evaluate the effect of sucrose addition to substrate and cryotube on the viability of Agaricus subrufescens cryopreserved at -20°C and at -75°C for one and two years. Zero, 10% or 20% sucrose was added to po...
متن کاملAssessment of Agaricus Bisporus S-II Extract as a Bio-Controlling Agent against Human Pathogenic Bacterial Species
Agaricus bisporus mushrooms are well known for their nutritional and medicinal values. A. bisporus is a source of protein (about 40% on a dry basis), ergosterol, several minerals, carbohydrate, and fat. The present study was conducted to investigate the effect of A. bisporus S-II extracts on human pathogenic bacteria in-vitro condition. Totally, thre...
